The symbols “>” and “<” before or after the reads point in the directions that the reads were generated. Nucleotide differences between the reference sequence and the aligned sequence are highlighted. In this case, a homozygous C>T transition that predicts a T355I amino acid substitution is supported by 42 reads. (B) Automated Sanger sequencing confirmed the C>T transition.
