Positive and negative controls included in the immunohistochemistry technique. a Positive control showing mast cells in an equine mast cell tumor. b Negative control using rabbit serum for the replacement of the primary antibody on a 3-day old limb wound. No cellular staining was observed but unspecific staining of epidermis, sebaceous and sweat glands, and hair follicles, was noted. An additional blocking step of the endogenous biotin was added to the protocol and limited the unspecific staining. The technique used the avidin-biotin-alkaline phosphatase method with Harris hematoxylin counter stain (× 200)
