TMUV infection induces XBP1 mRNA splicing and leads to p58IPK upregulation. a, XBP1 mRNA was amplified by RT-PCR and digested by Pst I. BHK-21 cells were mock-infected or infected with TMUV. Cells treated with 1 μM tunicamycin (Tu) for 12 h were used as a positive control. At the indicated times after infection, XBP1 mRNA was amplified by RT-PCR using XBP1-specific primers, after which the XBP1 fragment was subjected to Pst I digestion, and the products were separated by 1% agarose gel electrophoresis. b, p58IPK upregulation was detected by real-time RT-PCR. The Y axis shows the fold change of target gene expression in TMUV-infected cells, as determined by the comparative CT method. The data are expressed as the means±SD of results from three independent experiments. * p<0.05 vs. mock-infected cells. c, The protein level of p58IPK was examined by western blotting.
