From: Effects of glucose on lactose synthesis in mammary epithelial cells from dairy cow

Fig. 5

Protein kinase B knockdown decreased the expression of genes involved glucose transportation and lactose synthesis. Nonspecific scrambled siRNA (Scr) was used as a negative control in all experiments. a Relative mRNA levels of glucose transporter 1 (GLUT1), hexokinase-2 (HK2), β-1,4-galactosyltransferase 1 (β4GalT-I), and α-lactalbumin (α-LA) in siRNA-transfected dairy cow mammary epithelial cells. siRNA-transfected dairy cow mammary epithelial cells were cultured in DMEM with 12 mM glucose for 24 h. Cells were collected and the total RNA was isolated for gene expression analysis. The mRNA levels were measured by qPCR and normalized to β-ACTIN mRNA. b, c Western blot analysis (b) and quantification (c) of protein expression levels of GLUT1, solute carrier family 35 member A2 (SLC35A2), solute carrier family 35 member B1 (SLC35B1), HK2, and β4GalT-I in siRNA-transfected dairy cow mammary epithelial cells. siRNA-transfected dairy cow mammary epithelial cells were cultured in DMEM with 12 mM glucose for 24 h. Cells were collected for western blot analysis, and the protein levels were normalized to β-ACTIN. Data are shown as the mean ± SEM from 3 independent replicates. *P < 0.05; **P < 0.01
